Divergent splicing of paralogous genes – the intergenic regulatory loop of RPL22
Production of ribosomal proteins is strictly regulated to fulfil the actual needs of the cell. We have concentrated on a pair of paralogous genes, RPL22A and RPL22B, coding for Rpl22 protein in bakers yeast (Saccharomyces cerevisiae). In this case, the regulation is posttranscriptional – it depends on introns. Binding of Rpl22 protein hampers intron splicing, thereby preventing translation. We have analyzed the regulatory loop and demonstrated portability of the signals involved. Studies of RNA secondary structure of the intron hint at an RNA-switch mechanism. Binding of the protein would be the factor stabilizing splicing-restrictive conformation.
RPL22 regulatory loop seems to have evolved from a simple autologous feedback present e.g. in Kluyveromyces lactis. However, duplication of the gene and divergent evolution of the resulting paralogs gave the yeast more options for regulatory fine-tuning.
