Drugs and drug targets in the human malaria parasite Plasmodium falciparum: Artemisinin and beyond

Plasmodium falciparum is the most deadly species that causes human malaria, which worldwide leads to ~300 million clinical cases and about half a million deaths annually. Today artemisinins are the magic bullet against human malaria. But given the waning efficacy of artemisinins observed recently, and the resulting future danger of global artemisinin resistance, there is an urgent need to find new drugs that one day could replace artemisinins. Using a network based computational approach that we have pioneered we elucidate still unknown mechanisms of action and resistance mechanisms of key antimalarial drugs, such as artemisinin and chloroquine, and systemically explore new candidates in computational and in high-throughput studies.
